Constitutional Cytogenetics - Tissues

Referred out for testing

Source

Tissues (fetal, postmortem, stillborn, etc.)

For biopsies or surgical specimens from live patients, see Specimen Requirements.

Mnemonic CGLNON
Specimen Requirements

Collect tissue in a sealed sterile container with transport media or RPMI to keep moist.

DO NOT put tissue in formalin.  DO NOT freeze.

Fresh fetal tissue (muscle, pericardium, thymus, skin) and/or umbilical cord

Placental or products of conception are at a risk of maternal cell contamination and will be tested as a last resort if they are the only sample available.

Post-mortem specimens for Cytogenetics: please refer to Fetal Losses and Autopsy.

Fresh surgical specimens or skin biopsies from live patients: please call the laboratory at 403-955-7545 for specific instructions.

Specimen Handling

Submit fresh specimen as soon as possible to ACH Pathology for grossing. ACH Pathologist will submit specimen to ACH Cytogenetics.  Include completed requisition(s) and clinical history. Multiple specimens from a patient require separate requisitions for each specimen. When possible, please transport multiple tissue specimens from a patient at the same time.

If sample is taken after normal working hours (Monday - Friday 08:00 - 16:15), store at 4ºC and send the next working day.

Additional Information

Culturing of fibroblasts is no longer routinely performed. Instead, a DNA-based testing algorithm has been implemented (see link below). If fibroblast culturing is required for any purpose, please clearly indicate the need for cell culture.

The physician or pathologist must complete an AHS "Requisition for Constitutional Cytogenetic Services - Karyotype, FISH and RAD" for Rapid Aneuploidy Detection (RAD) testing only, or an AHS "Chromosomal Microarray (CMA) Requisition" (signed by an approved physician) for RAD and CMA testing. Requisitions and further instructions available under Cytogenetics tab at: www.albertahealthservices.ca/lab/page8667.aspx
