Specimen Requirements

1 x 4 mL dark green top sodium heparin or 1 x 4 mL dark green lithium heparin tube, NOT PST. 

Minimum: 3 mL whole blood

It is recommended that patient go to ACH for specimen collection.

Specimen Handling

DO NOT centrifuge. Send whole blood. Do NOT transfer to a secondary container.

DO NOT refrigerate or freeze; primary tube must remain at room temperature at all times.

Send a copy of the requisition with the specimen.

If not collected at ACH, transport whole blood to ACH at room temperature.

All samples SHOULD arrive before 14:00 and have been collected NO LONGER than 48 hours prior to arrival. If the sample needs to be stored following draw, store whole blood in original tube on benchtop at room temperature.

Additional Information

THIS IS NOT A DIAGNOSTIC TEST. This procedure involves the establishment and storage of an Il-2 stimulated T-lymphocyte culture for the purposes of Lysosomal Enzyme Testing.  Following T-lymphocyte clonal expansion, the cells are frozen down as pellets for testing.  This procedure cannot be used for continuous culturing or to re-grow a cell line.  This procedure is used to produce sufficient patient material for lysosomal enzyme testing of multiple enzymes, as clinically indicated (see below).

T-cell pellets are used for enzyme testing to investigate one or more of the following lysosomal storage disorders:
-alpha-Glucosidase (for Pompe Disease)
-alpha-lduronidase (for Hurler-Scheie Syndrome)
-Arylsulfatase A (for Metachromatic Leukodystrophy)
-Arylsulfatase B (for Maroteaux-Lamy Syndrome)
-beta-Galactosidase (for GM1 Gangliosidosis/Morquio B Syndrome)
-beta-Glucosidase (for Gaucher Disease)
-Galactocerebrosidase (for Krabbe Disease)
-Sphingomyelinase (for Niemann-Pick Disease Type A/B)
