FLT3/NPM1 Gene Mutations

Source Bone marrow preferred. Peripheral Blood or DNA also acceptable.
Mnemonic FLT3NPM1BM (Bone Marrow)
FLT3NPM1PB (Peripheral Blood)
FLT3NPMDNA (DNA)
Specimen Requirements

Tests can be performed on a Molecular Hematology specific sample (non-reflexive) or may be performed reflexively on certain residual bone marrow or peripheral blood specimens following Flow Cytometric analysis.

For reflexive testing:
NO separate specimen is required if specimen was collected in EDTA tube (lavender top).
Sample will be acquired from Flow Cytometry by Molecular Hematology.

For non-reflexive testing:
Bone Marrow Adults/Pediatric: Minimum 0.5 mL bone marrow in EDTA
Peripheral Blood: Adults - 1 x 4.5 mL EDTA or ACDA
Pediatric - minimum 1.0 mL in EDTA or ACDA
Send sample directly to Molecular Hematology.

Specimens collected in heparin (green top tube tubes) are UNACCEPTABLE for Molecular Hematology Assays.

Specimen Handling

Calgary zone: Send bone marrow/peripheral blood specimens with flow cytometry tests and FLT-3 to Flow Cytometry. Data enter only flow test mnemonics. If the Bone Marrow Pathology Requisition indicates Molecular Hematology or FLT3 BM or NPM1 and a separate sample is provided, order BMMolec and send to DSC Molecular Hematology.

Specimens for FLT3/NPM1 will be sent directly to Molecular Hematology from Flow Cytometry. Molecular Hematology will data enter the mnemonic FLT3/NPM1 as required.

Extra Regional: Send DNA extracted from bone marrow or peripheral blood to Molecular Hematology. Include special Third Party Requisition with sample. Bone marrow and peripheral blood may also be acceptable.

DSC Accession: after 16:30, store at 4oC. Do not centrifuge.

Additional Information Molecular investigations must be DEDICATED specimens to avoid sample contamination.

Peripheral blood specimens must be accompanied by a completed CLS Molecular Hematology requisition (REQ9053MH).

Bone marrow specimens must be accompanied by a completed Bone Marrow Pathology requisition (REQ9061BM).
