Specimens submitted to CLS from within Alberta: Mix well. DO NOT refrigerate, centrifuge or freeze. Deliver immediately to Flow Cytometry.

Flow Cytometry is used to measure surface CD107a expression on NK cells that have been stimulated to degranulate in response to incubation with a cell line, K562. NK cell granules (lysomes containing perforin and granzyme B) normally express CD107a and when their membranes fuse with the outer cell membrane upon exocytosis, CD107a then is present on the outer surface of the NK cell. This functional flow Cytometry assay is used to screen for familial hemophagocytic lymphohistiocytosis subtypes FLH3 (UNC13-D), FLH-4 (STX11), and FLH-5 (STXBP2) with defective NK cell exocytosis. The percentage of NK cells expressing CD107a after exposure to K562 cells is reported.
