Specimens submitted to CLS from within Alberta: Mix well. DO NOT centrifuge or freeze. Deliver immediately to Flow Cytometry.

Flow cytometric assessment of pSTAT5 is useful for investigation of a JAK3 gene mutation resulting in a primary immunodeficiency (SCID). This functional assay measures phosphorylation of STAT5 in CD3+4+ T cells after incubation and signaling by IL-6. Antibody to pSTAT5 is employed using an intracellular staining technique.
