Specimen Requirements

1 x 4 mL dark green top sodium heparin or 1 x 4 mL dark green lithium heparin tube, NOT PST.

Extra-regional*: 2 x 4 mL dark green top sodium or lithium heparin tubes, NOT PST.

Minimum: 0.3 mL whole blood

Pleural or cardiac effusion samples, or any other specimen types for the express purposes of tuberculosis testing, will NOT be accepted and will be disposed of upon receipt. For available tuberculosis testing, please contact ProvLab (403-944-1200).

Specimen Handling

DO NOT centrifuge; send whole blood. DO NOT transfer to a secondary container. DO NOT freeze (exception Extra-regional sites). Sample MUST arrive at ACH Biochemical Genetics Lab by 14:00.
Send a copy of the requisition with the specimen.

PSC and CRL: 
Transport whole blood on ice to DSC. Ensure tube is not in direct contact with ice pack so specimen does not freeze.

RRL:
Transport whole blood on ice to ACH. Ensure tube is not in direct contact with ice pack so specimen does not freeze.

Extra-regional:
Call ACH Biochemical Genetics Lab (403-955-7379) to make arrangements prior to shipment.  Ship 1 x 4 mL whole blood overnight on ice to ACH Biochemical Genetics Lab (ensure tube is not in direct contact with ice pack so specimen does not freeze). The second tube should be kept separate, centrifuged, plasma frozen and shipped on dry ice to accompany the whole blood sample.

Additional Information

Erythrocyte adenosine deaminase (ADA) and purine nucleoside phosphorylase (PNP) activities are measured to investigate the possibility of these underlying disorders in patients with an immunodeficiency.  Plasma ADA activity is also measured to aid in the initial diagnosis, as well as to monitor known ADA deficient patients on PEG-ADA therapy.

*It is preferable that extra-regional sites collect two separate blood samples for determination of plasma and erythrocyte adenosine deaminase activity: a whole blood sodium or lithium heparin, NOT PST sample for erythrocyte activity and a separate sodium or lithium heparin, NOT PST sample for the separation and storage of plasma.
